Assessment of DNA methylation profile during cryopreservation and long-term storage of germ cells

Abstract

В работе приведены результаты исследования влияния процесса криоконсервации на ДНК- метилирование гена H19 в сперматозоидах человека. Был применен комплексный подход к изучению данного процесса, включающий спермограмму, криоконсервирование, выделение ДНК, бисульфитную конверсию, капиллярное секвенирование по Сэнгеру и статистическую обработку данных. В результате пилотного эксперимента было показал, что процесс криоконсервации не оказывает влияние ни на качество ДНК, ни на профиль метилирования фрагмента гена H19. Однако данное утверждение требует продолжения исследования на большем числе образцов.

The paper presents the results of a study of the effect of the cryopreservation process on the DNA methylation of the H19 gene in human spermatozoa. A comprehensive approach was applied to the study of this process, including spermogram, cryopreservation, DNA isolation, bisulfite conversion, Sanger capillary sequencing and statistical data processing. As a result of the pilot experiment, it was shown that the cryopreservation process It does not affect either the quality of DNA or the methylation profile of the H19 gene fragment. However, this statement requires continued research on a larger number of samples.
