[2+5]-Cycloaddition of 2-arylidene-3-diazosuccinimides with nitriles — a novel approach to the synthesis of condensed azepines

Данная работа посвящена исследованию взаимодействия 2-арилиден-3-диазосукцинимидов с нитрилами и разработке на его основе нового метода получения бензазепинов. Показана применимость разработанной методики к синтезу ряда различных производных бензазепинов с хорошими и отличными выходами. Выявлены закономерности влияния периферических заместителей в диазосубстрате на ход реакции и выход целевых продуктов. Выяснено, что ограничением метода является наличие сильных электроноакцепторных групп в имидном фргаменте; в целом же реакция толерантна к широкому кругу заместителей. Предложен механизм реакции, включающий 1,7-электроциклизацию как ключевую стадию процесса и объясняющий обнаруженные закономерности. Также продемонстрирована возможность частичного восстановления полученных бензазепинов до дигидробензо[e]пирролоазепин-1,3-дионов с умеренными выходами.

This work is devoted to the study of the reaction of 2-arylidene-3-diazosuccinimides with nitriles and the development of a new method for preparation of benzazepines on the basis of this reaction. The applicability of the developed method to the synthesis of a number of different benzazepine derivatives with good and excellent yields has been shown. The pattern of peripheral substituents influence on the course of the reaction and the yield of the target products was revealed. It was found that the limitation of the method is the presence of strong electron withdrawing group in the imide fragment. In general, the reaction is tolerant to a wide range of substituents. The reaction mechanism has been proposed and includes 1,7-electrocyclization as a key step. The mechanism also explains the observed patterns. The possibility of partial reduction of the obtained benzazepines to dihydrobenzo[e]pyrroloazepine-1,3-diones was also demonstrated.
