Use of auxotrophic chlorella mutants for the determination of mutagenic activity of alkyl compounds of urea

Abstract

A modification of spot — testing procedure for evaluation of mutagenic effects of chemicals on green algae was proposed. The frequence of reversion from arginindependence to prototrophy was determined for number of auxotrophic mutants. The several substances could be tested on the same Petry dish with a layer of a suspension of auxotrophic cells in semisolid agar on surface of minimal media. The mutagens in paper disks were applicated together with trace of arginine (the growth factor). The latter one allows to pass the two sporulation of cells and enable the appearance of induced mutations in form of clones with changed phenotype (prototrophic). The experiments with synchronous culture of ARG-2 mutant give some indication on chromosomal localization of reversions from arginindependence to a prototrophic state

Предложена методика оценки мутагенной эффективности химических препаратов по частоте ревертирования ауксотрофных аргининзависимых мутантов хлореллы. В отличие от ранее применявшихся спот-тестов в предложенном способе на поверхность агаровой пластинки минимальной среды с клетками ауксотрофного штамма тестируемый препарат наносится одновременно со следовыми количествами ростового фактора. Предложенная методика позволяет снять эффекты задержки выщепления и фенотипической задержки проявления мутации.
