The study of regeneration process as a genetic character by using the method of tissue-culture of isolated organs

Abstract

For the stude of regeneration ability of diffefent forms of a genus Raphanus sativus var. radicola Pers. a method of tissue- and organ-culture of the seedling has been of seedling for explantats and also the method of culture of regenerants. Out of all the studied parts of the seedlings, the laminar part of the cotyledons appears to possess more ability for regeneration, which during the period of culture increased in size, restored its removed petiole and at the place of the wounds formed the callus or root. In the experiments a clear difference between the regenerants of the varieties "Virovsky belley" and "Sachs" in the degree of increase of size of the regenerants of the leaf-blades has been established. Differences in the size of regenerated petioles of the cotyledons and the callus on the wounded surfaces have also been noted.

С целью изучения регенерационной способности генетически различных форм одного вида (Raphanus vаг. Sativus V. radicola, Pers.) разработан способ культивирования органов и тканей проростков редиса: способ стерилизации семян, подготовки растения донора и деления проростка на эксплантанты, а также метод культивирования регенерантов. Наиболее регенерационноопоеобными из всех изучаемых частей проростка оказались листовые пластинки семядолей, которые в течение культивирования увеличивались в размерах, восстанавливали удаленный черешок, а на месте ранения формировали каллус или корешки. В опытах установлены четкие различия между регенерантами сортов Вировский белый и Сакса по степени увеличения в культуре размера регенерантов листовых пластинок, намечаются различия по размеру регенерировавшего черешка семядоли и размеру каллуса на раневой поверхности.
