dc.description.abstract | Амилоидами называют белковые фибриллярные агрегаты, сформированные за счет образования упорядоченных межмолекулярных β-складчатых листов. Изначально формирование амилоидных фибрилл ассоциировалось исключительно с патологиями. Образование патологических амилоидов связано с развитием таких социально значимых заболеваний как болезни Альцгеймера, Паркинсона и латеральный амиотрофический склероз. Однако с каждым годом появляется все больше данных о функциональных амилоидах. Такие белки были обнаружены у представителей самых различных таксонов, от бактерий до млекопитающих. Функциональные амилоиды обнаружены, в частности, у различных организмов в половых клетках и их оболочках. Амилоидные структуры были выявлены в оболочках яиц тутовых шелкопрядов, костистых рыб и млекопитающих. Также выделяют амилоидоподобные белки, формирующие агрегаты, которые проявляют некоторые из физико-химических характеристик амилоидов. Ранее в нашей лаборатории были получены данные, позволяющие выдвинуть гипотезу о существовании амилоидов в ооцитах курицы Gallus domesticus. Так, было показано, что амилоид-специфичный краситель Тиофлавин-S связывается с некими структурами в ооцитах, а также клетках фолликулярного эпителия на ранней стадии развития ооцитов. С помощью протеомного скрининга белков, формирующих SDS-устойчивые амилоидоподобные агрегаты, в ооцитах курицы был выявлен белок Vitellogenin-2 (VTG2), выполняющий функцию главного предшественника запасных белков яичного желтка, служащих основным источником энергии в ходе развития куриного эмбриона. Данная работа посвящена выявлению фрагментов белка VTG2, способных формировать детергент-устойчивые агрегаты в дрожжевой модельной системе. Согласно результатам предсказаний биоинформатического алгоритма «ArchCandy», последовательность VTG2 была разбита на три фрагмента, которые соответствуют его протеолитическим продуктам и содержат потенциально амилоидогенные последовательности. С помощью флуоресцентной микроскопии было показано, что все три фрагмента VTG2, слитые с YFP, способны формировать агрегаты в клетках дрожжей S. cerevisiae. Этот результат был подтвержден с помощью фракционирования дрожжевых клеточных лизатов и детекции исследуемых белков с помощью специфических антител. Проведенный биохимический анализ устойчивости фрагментов VTG2 к обработке 1% SDS показал, что только N-терминальный фрагмент VTG2 формирует частично детергент-устойчивые агрегаты. В целом, результаты, полученные в дрожжевой модельной системе, показывают, что различные последовательности белка VTG2 способны агрегировать. | ru_RU |
dc.description.abstract | Amyloids are fibrous protein aggregates formed by ordered intermolecular β-sheets. Formation of amyloid fibrils has traditionally been associated only with pathologies. The accumulation of pathological amyloid fibrils is associated with group of socially significant diseases including Alzheimer’s, Parkinson’s diseases and amyotrophic lateral sclerosis. Year after year, however, more and more data on functional amyloids appears. Such proteins have been found in a wide variety of taxa, ranging from bacteria to mammals. Functional amyloids have been discovered in particular in germ cells and its eggshell of various organisms. Amyloid structures are found in eggshells of silk moths, teleost fish, and mammalian oocytes. Besides, there are amyloid-like proteins that form aggregates exhibiting some of the physicochemical characteristics of amyloids. Our research team has previously obtained data that allowed to propose a hypothesis about the existence of amyloids in the oocytes of the chicken Gallus domesticus. It has been shown that amyloid-specific dye, Thioflavin S, stains follicular cells and significant part of the cytoplasm of chicken oocytes at the early stages of development. Proteomic screening of proteins that form SDS-resistant amyloid-like aggregates in chicken oocytes revealed Vitellogenin-2 (VTG2) protein. VTG2 is the main precursor of egg yolk storage proteins, which serve as the main source of energy during the development of the chicken embryo. This work is devoted to the identification of the VTG2 protein fragments capable of forming detergent-resistant aggregates in the yeast model system. The predictions of the ArchCandy bioinformatics algorithm have allowed to split the VTG2 sequence into three fragments that correspond to its proteolytic products and contain potentially amyloidogenic sequences. It was shown, using fluorescence microscopy, that all three VTG2 fragments fused with YFP are capable of forming aggregates in S. cerevisiae yeast cells. This result was confirmed by fractionation of yeast cell lysates and detection of the studied proteins using specific antibodies. The conducted biochemical analysis of the resistance of VTG2 fragments to treatment with 1% SDS showed that only the N-terminal VTG2 fragment forms partially detergent-resistant aggregates. In general, the results obtained in the yeast model system show that different sequences of the VTG2 protein can form aggregates. | en_GB |
