Influence of nutrient substrate composition and cultivation method on the biological activity and chromatographic profiles of extracts of the fungus Pestalotiopsis sp., isolated from Camellia sinensis leaves

Abstract

Грибы рода Pestalotiopsis известны как фитопатогены многих сельскохозяйственных растений. По данным литературы грибы этого рода являются продуцентами биологически активных веществ широкого спектра активности. Гриб Pestalotiopsis sp., использованный в работе, был выделен в чистую культуру из листьев чая (Camellia sinensis). Цель работы состояла в подборе питательных субстратов и способов культивирования для получения биологически активных метаболитов Pestalotiopsis sp., выделенного из листьев Camellia sinensis. Идентификация гриба было проведена с использованием морфолого-культуральных свойств и молекулярно-филогенетических данных. Культура гриба была получена на жидких средах M1D, ДМГ (среда на основе дрожжевого и мальтозного экстрактов), ЧАВ (среда Чапека с витаминами) и картофельно-глюкозном бульоне. В качестве твёрдых субстратов использовали рисовую, перловую и пшённую крупы. Антимикробную активность экстрактов из культурального фильтрата и мицелия гриба оценивали в отношении Pseudomonas syringae, Paenibacillus polymyxa и Candida tropicalis; фитотоксическую активность определяли методом листовых дисков на бодяке полевом (Cirsium arvense) и пырее ползучем (Elytrigia repens). Максимальный выход биомассы был отмечен при культивировании гриба на среде M1D и картофельно-глюкозном бульоне, минимальный - на среде ЧАВ. Было показано, что экстракты из культурального фильтрата гриба на средах ДМГ, M1D и картофельно-глюкозном бульоне, проявляют фитотоксическую и антимикробную активность в отношении тест-организмов. Экстракт из мицелия гриба на среде M1D также обладал фитотоксическими и антимикробными свойствами, в то время как экстракты из мицелия гриба на других жидких средах и твёрдых питательных субстратах были менее активны. При изучении метаболитного профиля экстрактов из культуры гриба было выявлено, что экстракты из культуры гриба на жидких питательных субстратах проявляют наиболее высокое разнообразие метаболитов. Экстракты из культуры Pestalotiopsis sp. на твёрдых средах содержали несколько общих метаболитов средней полярности. Таким образом, культивирование гриба на средах ДМГ, M1D и картофельно-глюкозном бульоне способствует образованию вторичных биологически активных метаболитов. Fungi of the genus Pestalotiopsis are known as phytopathogens of many agricultural plants. According to the literature, fungi of this genus are producers of biologically active substances of a wide spectrum of activity. The fungus Pestalotiopsis sp., used in the work, was isolated in a pure culture from the tea leaves (Camellia sinensis). The aim of the work was to select nutrient substrates and cultivation methods for the production of biologically active metabolites of Pestalotiopsis sp., isolated from the leaves of Camellia sinensis. Identification of the fungus was carried out using morphological and cultural properties and molecular-phylogenetic data. The culture of the fungus was obtained on liquid media M1D, YMG (medium based on yeast and malt extracts), Czapek's medium with vitamins and potato-glucose broth. As solid substrates used rice, pearl and millet. The antimicrobial activity of the extracts from the culture filtrate and the fungal mycelium was assessed for Pseudomonas syringae, Paenibacillus polymyxa and Candida tropicalis; phytotoxic activity was determined by the method of leaf discs on the fieldfeeding field (Cirsium arvense) and the more creeping creeping (Elytrigia repens). The biomass maximum was observed during cultivation of the fungus on medium M1D and potato-glucose broth, the minimum - on Czapek's medium with vitamins. It was shown that extracts from the culture filtrate of the fungus on media YMG, M1D and potato-glucose broth, show phytotoxic and antimicrobial activity against test organisms. The mycelial extract of the fungus on M1D medium also had phytotoxic and antimicrobial properties, while extracts from mycelial fungus on other liquid media and solid nutrient substrates were less active. When studying the metabolic profile of extracts from the fungus culture, it was found that extracts from the fungus culture on liquid nutrient substrates exhibit the highest variety of metabolites. Extracts from the culture of Pestalotiopsis sp. on solid media contained several common metabolites of medium polarity. Thus, cultivation of the fungus on YMG, M1D and potato-glucose broth promotes the formation of secondary biologically active metabolites.
